Therapeutic Application

Pulsed LIFU (0.5MHz, <10% duty cycle) activates astrocytic TRPA1 mechanosensitive channels -> Ca2+ influx -> Best1-mediated glutamate release (gliotransmission) -> extrasynaptic NR2B-enriched NMDA receptors -> neuronal depolarization. Distinct: (1) acoustic not EM, (2) astrocyte-first not neuron-first, (3) gliotransmission not synaptic vesicle, (4) extrasynaptic NMDA pool pharmacologically distinct from synaptic. Confirmed: Gu & Han 2019, Current Biology.
